LDSC++: a new method to enable more powerful multivariate GWAS analyses

Description

Linkage Disequilibrium (LD) score regression is a technique used to evaluate the genetic heritability and covariance of polygenic traits based on data from genome-wide association studies (GWAS). This method is often used in combination with multivariate methods like Genomic Structural Equation Modelling (SEM). In Genomic SEM, the genetic covariance structure and the regression intercepts of LD score regression are analysed to account for sample overlap. LDSC++, a novel implementation of LD score regression, aims to improve the accuracy of this method. Specifically, LDSC++ focuses on enhancing the regressions of bivariate trait combinations. LDSC++ builds upon the previous multivariate interpretation of LD score regression in Genomic SEM to improve the accuracy and utility of this important technique.
