Research interests

Dr Kudsiova is currently interested in studying the fate of non-viral gene delivery vectors such as lipid:DNA complexes inside cells. By fluorescently labelling each component of the gene delivery vector with various fluorescent labels, it is possible to track the intracellular fate of the DNA and the lipid over a period of time using confocal fluorescence microscopy.
Recent research has focused on using cationic lipid:DNA complexes incorporating a synthetic bifunctional peptide, pep6, consisting of a Lys16 domain (to bind DNA), and a cyclic domain (CRRETAWAC) known to bind specifically to the cell surface protein, a5b1 intergrin. Incorporation of this ligand was found to greatly improve the transfection efficiency of various lipids in a range of cell lines, and greatly enhance the transport of DNA into the cell nucleus. This work is carried out in collaboration with Dr Hailes and Tabor of the Chemistry Department UCL, and Prof. Ng and Dr. Keppler of the Randall Division of Cell & Molecular Biophysics, KCL.

Other areas of interest:

  • Investigating the intracellular uptake and trafficking pathways of lipid:DNA complexes by fluorescently labelling certain cellular compartments and using certain endocytotic pathway inhibitors
  • Investigating the physicochemical properties and the phase behaviour of cationic lipid:DNA complexes over a pH range that mimics endosomal conditions
  • Investigating the effect of chitosan and chitosan-DNA complexes on epithelial permeability across epithelial cell monolayers
  • Structural studies on ternary complexes of DNA-zwitterionic polymer-Ca2+ complexes
