Clusterin is both a genetic risk factor and a potential biomarker for AD. Its role in disease however is uncertain; as a pleiotropic protein it might plausibly be involved in lipid metabolism, in innate immunity or in amyloid trafficking. Clusterin is also part of the regulation of cell survival and we recently showed that Abeta induces changes in the cellular distribution of clusterin and this in turn induces DKK1 expression and a pathway mediating neurotoxicity. An expression signature indicative of this pathway is present in brain of animals with amyloidopathy but not tauopathy and in brains of people with AD but not FTD.
